The tissue should be attached with a tissue string on the transducer at the top and the hook at the bottom of the water cell, with the tissue in the middle
Most importantly, before the experiment starts, the scientists have to make sure that the tissue has a pre-load, i.e. that a small but significant tension is applied by the knobs.
The string should be in tension, as before starting the real measurement some level of pre-tension is needed by the tissue to react significantly and repeatably to the stimuli which will be applied. If the string is loose, this may mean that either the tissue is not well mounted to the transducer at the top or to the water cell hook at the bottom
